| Gram's staining techique |
This particular experiment uses the Gram's Stain method to
categorize bacteria into Gram's positive bacteria and Gram's
negative bacteria.
|
|
1.Collect plague and smear evenly on slide.
|
2.Air dry smear and then perform heat fixation. |
|
|
|
|
3.Crystal Violet Stain (1 minute).
|
4.Wash away stain and dry slide.
|
|
|
|
|
5.Apply Gram's iodine for 1 minute.
|
6.Wash away Gram's Stain.
|
|
|
|
|
7.Wash with 95% percent alcohol until alcohol runs clear.
|
8.Wash away alcohol with water, dry slide.
|
|
|
|
9.Counterstain with safranin for 45 seconds.
|
10.Wash away safranin with water.
|
|
|
|
|
11.Blot dry with bibulous paper, then observe.
|
