Honolulu Technique
The Honolulu Technique, developed by Dr. Teruhiko Wakayama and Ryuzo Yanagimachi from the University of Hawaii, involves inserting a nucleus into an enucleated egg. The success of this technique resulted in further research into exactly how an egg reprograms a nucleus.
First, the nucleus from a somatic cell must be removed. This nucleus is then transferred into an egg cell that has had its nucleus removed. The resulting egg is placed in a chemical solution and cultured. Unlike the Roslin technique used to create Dolly, no culturing was done on the cells prior to fusion. The cells accept the new nucleus in about an hour. After an additional five hours, the resulting egg cell is placed in a chemical culture to initial cell growth. As in other techniques, the developing embryo is then implanted into a surrogate for development.
